Abstract

Keberadaan Cendawan Di Daerah Rhizosfer Dipengaruhi Oleh Berbagai Faktor, Salah Satunya Adalah Bahan Organik Tanah. Penggunaan Bahan Organik Berupa Pupuk Kandang, Limbah Udang Dan Air Kelapa Pada Budidaya Sawi (Brassica Juncea L.) Belum Di Ketahui Pengaruhnya Terhadap Keragaman, Kelimpahan Dan Aktifitas Fisiologi Cendawan Di Daerah Rhizosfer, Oleh Karena Itu Penelitian Ini Bertujuan Untuk : 1) Mengetahui Keragaman Dan Kelimpahan Cendawan Daerah Rhizosfer Tanaman Sawi (Brassica Juncea L.) Dengan Pemberian Limbah Udang Dan Air Kelapa 2) Mengetahui Aktifitas Fisiologi Cendawan Yang Diperoleh. Penelitian Ini Terdiri Dari Beberapa Kegiatan Yaitu Pengambilan Sampel Tanah, Isolasi Cendawan, Purifikasi Cendawan, Pengamatan Makroskopis, Pengamatan Mikroskopis, Uji Aktifitas Melarutkan Fosfat, Uji Aktifitas Menghidrolisis Protein, Uji Patogenisitas, Dan Uji Antagonisme Terhadap Fusarium Oxysporum.
Hasil Penelitian Menunjukkan Kelimpahan Dan Keragaman Koloni Cendawan Yang Berbeda Pada Tanah Dengan Bahan Organik Yang Berbeda. Pada Sampel C1 Yaiitu 50 Koloni, Sampel C2 Yaitu 25 Koloni, Sampel C? Yaitu 9, Sampel C4 Yaitu 7 Koloni, Selanjutnya Diikuti Pada Sampel C5 Yaitu 6 Koloni Cendawan. Koloni Cendawan Yang Diperoleh Menunjukkan Perbedaan Dalam Hal Karakterisasi Pada Sampel C1 Yaitu Sebanyak 12%, C2 Yaitu Sebanyak 38%, C3 Yaitu Sebanyak 22%, C4 Yaitu Sebanyak 28%, Dan Diikuti Pada Sampel C5 Yaitu Sebanyak 16%. Aktifitas Melarutkan Fosfat Menunjukkan Kemampuan Isolat Pada Sampel C1 Yaitu 5 Isolat, Diikuti Oleh C2 Yaitu 3 Isolat, C4 Yaitu 1 Isolat, Dan C5 Yaitu 1 Isolat. Sedangkan Aktifitas Menghidrolisis Protein Diperoleh Pada Sampel C1 Yaitu Sebanyak 3 Isolat, Dan Diikuti Oleh C2 Yaitu Sebanyak 4 Isolat. Uji Antagonisme Terhadap Fusarium Oxysporum Menunjukkan 5 (Lima) Isolat Cendawan Mampu Menekan Pertumbuhan F. Oxysporum, Dan Sembilan Isolat Cendawan Nonptogenik Termasuk Dalam Genus Yaitu Penicillium Sp., Peacilomyces Sp., Phiophora Sp., Dan Scopulariopis Sp.
Berdasarkan Hasil Penelitian Dapat Disimpulkan Sebagai Berikut : 1) Kelimpahan Koloni Cendawan Tertinggi Diperoleh Pada Sampel C1 Yaitu Sebanyak 50 Koloni Dan Kelimpahan Koloni Cendawan Terendah Diperoleh Pada Sampel C5 Yaitu Sebanyak 6 Koloni. Sedangkan Keragaman Karakterisasi Tertinggi Diperoleh Pada Sampel C2 Yaitu Sebanyak 38%, Diikuti Dengan C4 Yaitu Sebanyak 28% Dan Keragaman Karakteriasi Terendah Pada Sampel C1 Yaitu Sebanyak 12%. 2) Aktifitas Melarutkan Fosfat Tertinggi Diperoleh Dari Sampel C1 Dan Sampel C5 Dengan Diameter Zona Bening Terbentuk 2,6 Cm, Sedangkan Aktifitas Proteolitik Tertinggi Diperoleh Dari Sampel C1 Dan Sampel C2 Dengan Diameter Zona Bening Terbentuk 1,8 Cm. Sembilan Isolat Cendawan Nonptogenik Termasuk Dalam Genus Yaitu Penicillium Sp. (3 Isolat), Peacilomyces Sp. (3 Isolat), Phiophora Sp. (2 Isolat), Dan Scopulariopis Sp. (1 Isolat). Penelitian Ini Menghasilkan Isolat Cendawan Yang Memiliki Potensi Untuk Digunakan Sebagai Pemacu Pertumbuhan Melalui Serangkai Penelitian Selanjutnya.

The Existence Of Fungus In Rhizosphere Area Are Influenced By Several Factors, One Of Them Is Soil Organic. The Use Of Organic In The Form Of Manure, Water Fertilizer And Also Coconut Water On Cultivation Of Mustard (Brassica Juncea L.) Is Not Knowing As The Effect Of The Diversity, Abundance And Also Physiological Activity Of Fungus In The Rhizosphere Area. Because Of That The Aims Of This Research Are: 1) To Know The Diversity And The Abundance Of Fungus Mustard Plant (Brassica Juncea L.) With Giving Waste Fertilizer Of Shrimp And Coconut Water 2) To Know The Physiological Activity Of Fungus Is Obtain. This Research Consists Of Several Activities, As Follows : Taking The Soil Sample, Fungus Isolation, Fungus Purification, Macroscopic Observation, Microscopic Observation, Phosphate Dissolving Activity Test, Protein Hydrolyzing Activity Test, Pathogenicity Test, And Antagonism Test On Fusarium Oxysporum. The Results Of This Research Showed That The Abundance And The Diversity Of Fungus Colonies Which Are Different In Soil And Organic Material. On C1 Sample Are 50 Colonies, C2 Sample Are 25 Colonies, C3 Sample Are 9 Colonies, C4 Sample Are 7 Colonies, Next On C5 Sample Are 6 Colonies Of Fungus. The Colonies Of Fungus Are Obtained Showed The Differences Of Characterization On C1 Sample Are 12%, C2 Are 38%, C3 Are 22%, C4 Are 28 % And Followed By C5 Sample Are 16%. The Activity Of Dissolving Phosphate Showed That The Isolate Ability On C1 Sample Are 5 Isolates, Followed By C2 Are 3 Isolates, C4 Are 1 Isolate, And C5 Is 1 Isolate. Meanwhile, The Activity Of Hydrolysis Protein Is Obtained In C1 Sample Are 3 Isolates, And Followed By C2 Are 4 Isolates. An Antagonism Test On Fusarium Oxysporum Showed That 5 (Five) Fungus Isolates Are Able To Press The Growth Of Fusarium Oxysporum, And Nine Isolates Of Non-Pathogenic Inside On The Genuos Of Penicillium Sp., The Peacilomyces Sp., The Genuos Of Phiophora Sp., And Genuos Of Scopulariopis Sp. Based On The Results Of The Research, Could Be Concluded As Follows: 1) The Abundance Of The Highest Colonies Fungus Are Obtained On The C1 Sample That Are 50 Colonies And The Abundance Of The Lowest Colonies Fungus Are Obtained In C5 Sample Are 6 Colonies. Meanwhile The Highest Of The Diversity Characterization Was Obtained In C2 Sample Which Are 38%, Followed By Sample Which Are 28% And The Lowest Of The Diversity Characterization In C1 Sample Which Are 12%. 2) The Highest Of Phosphate Dissolving Activity Was Obtained From C1 Sample And C5 Sample With Diameter Zone Is 2.6 Cm. Meanwhile The Highest Of Proteolytic Activity Was Obtained From C1 Sample And C2 Sample With Diameter Zone Is 1.8 Cm. Nine Isolates Of Nonptogenic Fungus Included In Genus, They Are Penicillium Sp. (3 Isolates), Peacilomyces Sp. (3 Isolates), Phiophora Sp. (2 Isolates), And 1 Genus Of Scopulariopis Sp. This Research Produces The Isolates Fungus That Has Potential To Used As A Growing Spurt Through A Series Of The Subsequent Research.
