Abstract
Penelitian Ini Bertujuan Untuk Mengetahui Keragaman Cendawan Rizosfer Pada Pertanaman Padi. Perbanyakan Dan Pengujian Cendawan Rizosfer Dilaksanakan Dilaboratorium Perlindungan Tanaman. Fakultas Pertanian. Universitas Borneo Tarakan. Pada Bulan September 2017 Sampai Januari 2018. Alat Dan Bahan Yang Digunakan Antara Lain Disposible Petridish, Tabung Reaksi, Sendok Pengaduk, Cawan Petri, Laminar Air Flow, Erlenmeyer, Bunsen, Micro Pipet Dan Autoclafe. Sedangkan Bahan-Bahan Yang Digunakan Adalah Plastik Wraping, Aluminium Foil, Alkohol 70%, NaOCl, Spiritus, PDA, Benih Padi Varietas IR 64 Dan Benih Mentimun Varietas Mercy. Pelaksanaan Penelitian Dimulai Dengan Pengambilan Sampel Tanah, Isolasi Cendawan, Identifikasi, Mengamati Pertumbuhan Cendawan, Uji Aktifitas Proteolitik Serta Pengujian Patogenitas Cendawan.
Berdasarkan Identifikasi Telah Ditemukan Lima Jenis Cendawan Yang Berhasil Diisolasi Pada Areal Persawahan Di Tanah Masam, Ketujuh Jenis Cendawan Ini Antara Lain Pythium Sp, Trichoderma Sp, Phytophthora Sp, R. Solani, Rhizoctonia Sp, Dan Penicillium Sp. Hasil Pengamatan Karakteristik Fisiologi Menunjukkan Bahwa Tidak Semua Cendawan Memiliki Kemampuan Mendegradasi Protein, Diperjelas Pada Tabel 4 Pengujian Proteolitik Didapatkan Hasil Dari Ketujuh Isolat Cendawan Hanya Cendawan Dengan Kode Isolat C7 Yang Memiliki Zona Bening Yang Menandakan Telah Terjadinya Proses Pendegradasian Protein. Hasil Uji Patogenesitas Ditemukan Dua Jenis Cendawan Yang Bersifat Non-Patogen Dengan Kode Dan Persentase Tumbuh Benih Yaitu C2 (45%), C7 (75%), Sedangakan Lima Jenis Cendawan Yang Bersifat Patogen Yaitu Cendawan Dengan Kode Dan Presentase Tumbuh Benih, Adalah C1 (0%), C3 (0%), C4 (0%), C5 (0%) Dan C6 (0%).
Kesimpulan Dari Penelitian Ini Adalah Ditemukannya Cendawan Yang Bersifat Non-Patogen Antara Lain Trichoderma Sp Dan Penicillium Sp, Sedangkan Yang Bersifat Patogen Adalah Pythium Sp, Phytophthora Sp, R. Solani, Dan Rhizoctonia Sp. Serta Pada Uji Proteolitik Hanya Cendawan Dengan Kode Isloat C7 (Penicillium Sp) Yang Memiliki Kemampuan Dalam Mendegradasi Protein.
This Study Aims To Determine The Diversity Of Rhizosphere Fungi In Rice Plantations. Propagation And Testing Of The Rhizosphere Fungus Were Carried Out In The Plant Protection Laboratory Faculty Of Agriculture. Borneo Tarakan University. From September 2017 To January 2018. Tools And Materials Used Include Disposable Petri Dish, Test Tubes, Stirring Spoons, Petri Dishes, Laminar Air Flow, Erlenmeyer, Bunsen, Micropipette And Autoclave. While The Materials Used Were Wrapping Plastic, Aluminum Foil, 70% Alcohol, NaOCl, Spirits, PDA, IR 64 Variety Of Rice Seeds And Mercy Varieties Cucumber Seeds. The Research Began With Soil Sampling, Fungus Isolation, Identification, Observing Fungus Growth, Proteolytic Activity Tests, And Fungal Pathogenicity Testing. Based On The Identification Of Five Types Of Fungi Which Were Successfully Isolated In Rice Fields In Acid Soil, The Seven Types Of Fungi Included Pythium Sp, Trichoderma Sp, Phytophthora Sp, R. Solani, Rhizoctonia Sp, And Penicillium Sp. The Observations Of Physiological Characteristics Showed That Not All Fungi Could Degrade Proteins, It Was Clarified In Table 4 Proteolytic Testing Showed That The Seven Fungus Isolates Were Only Fungi With A C7 Isolate Code That Had A Clear Zone Indicating A Protein-Degrading Process. The Results Of The Pathogenicity Test Found Two Types Of Fungi That Were Non-Pathogenic With The Code And Percentage Of Seed Growth, Namely C2 (45%), C7 (75%), While Five Types Of Pathogenic Fungi Namely Fungi With Code And Seed Growth Percentage, Were C1 (0%), C3 (0%), C4 (0%), C5 (0%) And C6 (0%). The Conclusion Of This Study Is The Discovery Of Non-Pathogenic Fungi Including Trichoderma Sp And Penicillium Sp, While Those That Are Pathogenic Are Pythium Sp, Phytophthora Sp, R. Solani, And Rhizoctonia Sp. As Well As The Proteolytic Test Only The Fungus With C7 Isolate Code (Penicillium Sp) Can Degrade Protein.
